Great question- personally I enjoyed The Adventures of Kavalier and Clay- I think it was the book before YPUnion and gained Chabon the most popularity, but if YPU is interesting you then read on.

I just loved Wonder Boys, the story is so original and funny. Although you have to read the novel before you watch the movie. The movie is okay, but as always gives away everything, plus you should have the chance to imagine these wonderful characters in your head.

I tried reading Kavalier & Klay, I stopped reading after the first part.. it's just really overwritten I think.
Read The Mysteries of Pittsburgh first--it's Chabon's take on The Great Gatsby. Then read Wonderboys. Then read K & K. You can see Chabon's progression if you do it this way.
I have to disagree about K & K being overwritten. I'm amazed by the guy's sentences and detail. Usually the rule goes: write the fantastical stuff using simple language and the mundane stuff with verbal acrobatics. He goes after it all; there's amazing action, amazing detail, and amazing sentences all packed into one.
I suppose you have to enjoy the verbal indulgence.
